There is a delayed adverse health effect in children surviving cancer and chemotherapy .
The early effects of doxorubicin-induced cardiotoxicity have been described, but late effects (8-20 yr) are unknown. This pre-clinical study carried out with Wistar rats sought to understand if doxorubicin-induced “late-onset cardiotoxicity” is the result of mitochondrial dysfunction leading to cell failure and death. Results indicate that 6 months of doxorubicin injection significantly increased myocardial mitochondrial DNA damage (mtDNA), but the same damage was not found in c-kit left ventricular (LV) cells. Within the LV there was significant increase in biomarkers of senescence as well as histamine content.
Taken together the results indicate that doxorubicin therapy is associated with significant damage to the cardiomyocyte population.
